CLL14: In a randomized (1:1), multicenter, actively controlled, open-label, phase 3 trial (CLL14), VEN+G was studied against GClb in 432 patients with previously untreated CLL with comorbid medical conditions (total CIRS score >6 or CLcr <70 mL/min). The primary endpoint was progression-free survival (PFS). See full study design.1,2
reduction in risk of progression or death vs GClb (HR=0.33; 95% CI: 0.22-0.51 [P<0.0001])
After a median follow-up of 28 months (range: 0-36 months):
5-year follow-up analysis of INV-assessed PFS (overall follow-up of 74 months)5,10†
At the time of the analysis†:
71% of patients in the VEN+G arm had not received subsequent treatment (vs 42% of patients in the GClb arm).
*Number of events based on earliest event of disease progression or deaths without disease progression due to any cause.1
†Based on data as of clinical cutoff date of November 8, 2021; time of analysis was not pre-specified.
‡Time to next treatment was defined as the time from randomization to start of a new CLL therapy or death.
1L=first-line; CI=confidence interval; CIRS=Cumulative Illness Rating Scale; CLcr=creatinine clearance; CLL=chronic lymphocytic leukemia; HR=hazard ratio; IRC=Independent Review Committee; GClb=GAZYVA + chlorambucil; PFS=progression-free survival; SLL=small lymphocytic lymphoma; VEN+G=VENCLEXTA + GAZYVA.
